Default GS450H Tire Sizes

Stock is 245/40/18, will 245/45/18 fit on the car? Will the ride feel more smooth and less bumpy?

Difference is about half an inch or a little less. Ideally it should but seeing you have the Hybrid, I'd say just stick with the same size. That way your odometer and mpg wont change too much. More or less I would recommend with picking a better tire or a softer tire depending on what tire you have on them right now. The only way to solve your second question more or less is to go to the 17" wheels.

Or, it could be your suspension getting worn and is just time to replace.

I think your idea has merit. The larger tire will only impact your odometer will only be about 1 mile per hour off at 60. Also, your gas mileage should increase, if anything, with the larger tire diameter. I am doing this exact swap as we speak.
