water in wheel well

I have a 2996 ES 330. Water collects in the right rear wheel well. What causes this? Should I drill a hole to let it drain?

Dont drill a hole, theres a plug. Also, probably your taillight seal is leaking, or the sunroof drain tube came out of the socket, but you would be able to see that
