Need help with mom's 99 ES300 - Cylinder Misfires, VVT Sensor, and A/F Sensor issues

P0305 Cylinder 5 Misfire
P0306 Cylinder 6 Misfire (new)

Going to drive around a little more to see what codes stablize.
I would swap ignition coils, clear the codes and see if they change, before buying new ones.
Its common for the oil control valves to fail around the same time/mileage.
When I replaced mine I only received the misfire 2, 4, 6 and random misfire codes and vice versa for bank 1. No vvti code.
Look up es300 denso ignition coils on Amazon, good replacement.

It would be easy to swap the cylinder 6 coil with the #2 coil. I'd do that first and see it the misfires follows the coil. It's also possible the bank 2 OCV is bad or clogged up. You might want to consider cleaning the OCV filters and pulling the new OCV and see if it got fouled. I had one OCV go bad. Immediately after I replaced it the other went bad. Also with the flashing CEL it is not a good idea to be driving the car. Damage could occur.

Are there any diagrams of where the coils and #s are on the ES300, and are they easy to get to and swap/replace? Thanks!

pop the hood. With you facing the engine, from left to right as follows:
front: 2, 4, 6
back: 1, 3, 5

Here's a photo from the front of the car. The engine's decorative cover has been removed. See the three black things at the bottom of the photo. Those are the coils, from left to right 2, 4, 6. Disconnect the electrical connectors. Unscrew the bolt. Pull them out. The other coils are behind the engine plenum, closer to the firewall. They are a lot harder to access. Some contortionists have been able to remove the bank 1 coils and spark plugs without removing the plenum. I am not in that club.

Thanks for the pics. Makes it a lot easier to understand. Haven't gotten around to this yet, but read in another post that:
the coils are paired 1-4, 2-5, 3-6 meaning logically if you pull mis-fires on associated cylinders, your coil was trashed. Otherwise, you just 99.9983746% of the time needed new plugs, and you just let someone take you to the cleaners)

Since I'm pulling P0300, P0305, P0306 and 5-6 is not associated, does that mean it's most likely spark plugs? Can spark plugs cause the same symptoms as coils? (rough idle, constant vibration). I think spark plugs are pretty cheap and I can just get them from the local auto store.

Bad spark plugs could be causing this. When was the last time you changed spark plugs? I'd bet they haven't been changed since your car was new. You probably need new ones anyway. Buy the iridium NGKs, about $8 on Amazon. They'll last 100,000 miles.

My ES300 is a 2000 but the igniter coils go left to right. Front bank 2 (remember the motor is sideways)
Passenger lt #2, #4, #6
Bank 1 is back same order #1, #3, #5.
Hardest part is #3 b/c under the dohc manifold. It can be done without removing, use a swivel head rachet/ socket wrench and make sure the engine is cool. #10 coil bolt, #5/8 plug. Need a deep well and a 3" or 6" inch extension bar.
Clear codes and dive at least 30-50 miles with several stops and cut off restarts to reset computer recognize part. Idk why but it has to relearn some sensors.
