I just got my 05 LS430 w/ 46k miles. I had it inspected before I bought it and the tire wear is uneven. Most wheels are around 4/32nds and one is at 7/32nds. I had the TPMS light go off on the long drive home from Dallas where I bought it. I found the reset button and the light was off for another 80 miles or so but it came on last night driving home from work. Initially the tires were at 50psi and the spare was at 14, I aired them all up.

If the sensor works on change in wheel speed then does the LS430 need all tires replaced each time one needs to be replaced?

LS430 should be a direct system... meaning it checks the air pressures individually through a wireless transmitter...

you need to go check each of the pressures yet again... a small difference in tread height should not affect a non-AWD vehicle...

I would say adjust the tire pressure and make sure you get an alignment after the next set get installed.
