I would call Lexus dealership -> parts and ask them. They keep track of latest recommended parts/fluids. For my RX300, I get the Toyota Red coolant and mix it with distilled water [1 : 1.25 water]. There is a chart for the mix ratio on the bottle. More water improves heat transfer, but you risk freezing.
If you want to do a bit more, you can add "water wetter" by Redline. This reduces the water tension and improves coolant adhesion to engine and radiator wall and improves heat transfer. Less hot spots .. if you believe the literature.

How many miles are on your 2007? If low, say under 60K, this makes sense to flush and fill. However, if you have high mileage, say over 80K, you might want to hold off as you will soon be needing to have your timing belt replaced and you might as well do all that at the same time.
Also, keep in mind salimshah's comment to use only distilled water. Anything else may contain mineral deposits which could lead to problems later.

If you are topping off in the radiator it self, reaching for water or conc or premix, all would work as there is enough volume of the mix. If you are topping off the overflow, water would be bad as if temp falls below freezing it will freeze and expand as solid. Conc would work as the heat exchange is with the environment and not the engine.
Depending on how cold it gets, one can adjust the mix ratio. In central Texas, I use 1gal bottle of conc and add water to fill the system uo [that works out to be 1:1.25]
I hope it demystifies the issue.
[I substitute H2O for water]

If you plan to DIY, read Lexmex's instructions.

It is great coolant. If you want to do anything look for a better thermastat but not sure if they sell one for the RX.
DO NOT mix RED and GREEN COOLANT. Toyota is red coolant so use Toyota. Do not mix some green Prestone or something with the red.

Go to parts and ask if they have both [premixed or/and concentrate]. They will even tell you how many bottles you must purchase for a drain and fill (if you go to Lexus). Price them out and add the cost of distilled water for dilution. If they have only one, then they will make the decision for you.
You can eye ball the mixing or use a measuring cup. If your mix is off by couple of drops, it would not be a disaster. The procedure to drain is 10x more complex than mixing two liquids.

this is just huge for me. i did not know that you need to add water in your coolant system.
all i do is buy antifreeze/coolant and then pour it in when it was low.
i never add any water, have been doing this for 3~4 years now. ( not on the lexus )

Just follow the instructions on the bottle and you will do fine.
I am not saying this as a jest or to mess-up your vehicle.
