Selecting to return to Home on the Enform Direction Assistance to get me out of a big city. Once out of the city how do I stop cancel the direction assistance. I do not want to delete the destination just stop the voice assistance.

Several ways. One way is Menu / Setup / Voice / Voice Volume / set it OFF

Another way is click "Route" on the bottom of the map screen and then click "Pause Guidance". You can then resume whenever you wish in the same menu.

If you are concerned with "deleting" the destination, it will NOT be permanently deleted (at least on our '08 400h & '09 TCH.)

That terminology is a pet peeve of mine. Choosing "delete" when working with computers almost always means whatever it is will be "gone" from a database or storage medium. In this area of the Toyo NAVs, it just means that the current navigation routing process will be stopped. The actual destination remains in either "home" or "previous destinations" areas.

Instead of "delete this destination," the choice should be "stop current nav routing" or something like that. Dunno if this applies to 3rd gen RX NAVs or not, but I suspect it does. Toyo is always behind the curve on this kinda stuff.

FWIW even though it's off-topic from the OP's original question, I can easily delete previous destinations in my 2013 from Menu / Destination / Previous Destinations / Delete ... and then select an individual address or "Select All". I don't remember how I did the same thing in my 2006 RX400h as I've been without it now nearly 18 months, but am sure my previous destinations were deleted in the steps I took prior to my trade-in. I double-checked myself on all that.
Personally, I find my Lexus keeping my most recent previous destinations a huge plus. It's what I use most of the time to gain the advantage of automatic reroute based on current traffic jams.

On the whole "meaning of delete" point, I get what you're saying as my professional heritage is as a software programmer long ago, but in the context of Lexus Nav, I've learned "delete" when it comes to an active route means in their implementation "to the next destination"... i.e. If I have only a single destination active, the routing is ended (and saved in previous destinations like I would expect it to), but if I have an intermediate destination in my current route, only the part of the route I've selected is "deleted" from the routing underway. I don't find it a big deal, and guess I've just learned what they mean by "delete" in this context of the Nav.

Turn up the volume, LOL! I rarely hear nav lady when I'm driving. That's one thing Lexus mysteriously changed on the 3rd gen, no longer muting the stereo when nav lady speaks. At least the turns display in HUD. And press the map button to repeat directions if you miss them.
