When I got my 330 at 73K I had to repair the stuff that had probably made the original owner trade it in the first place. The wholesaler I got it from had replaced the battery and a coil which was no doubt a prime motivator and the rear tires were cupped and sounded for all the world like both rear wheel bearings needed replacement. It also had a leaking steering rack and needed rear brakes.
It was just an '05 330 that needed some midlife love. I had salvaged the new tires and 18" wheels from my totaled RX300 so that solved the rear end noise. I took the bad rear brakes as an opportunity to replace all four rotors with new drilled ones combined with Akebono ceramic pads all around and installed a rebuilt factory rack.
We replaced all the fluids and aligned all four wheels and did a detail. All the TSBs are up to date. I've done quite a few mods but those are for fun and aren't part of breakage and maintenance.
Except for the flurry of repairs when I first got it there have been no other incidents.

Overall been happy with the RX. Got the major repairs under TSB's and extended warranty took care of the rest.
Forbes lists Lexus' 3 cars on the top 10 (maybe even 4) most reliable cars in terms of repairs.
I saw LS460 (#1), ES350, GS350 on the list.

In addition to my earlier post in the thread, I have to add this one..
2006 RX400h, hybrid transaxle (err..we call it a transmission, Lexus calls it a transaxle) replaced at 155000 KM as it was leaking coolant. Roughly a $5000 part with labour on top of that. Fortunately for me, it was 100% covered under the extended warranty. My earlier post was the hybrid inverter, a $7000 part, also covered but under the Hybrid Warranty, not the extended warranty.

My 2007 RX 350 has been rick solid. Only issue that was non normal maintenance due to normal wear and tear (e.g. brake pads, tires, battery, etc) was having to change the rear hatchback motor and pistons. They were going up half way then would stop. Dealer lubricated for a few visits then replaced the parts listed under warranty. We are at 71,000 miles at the car has lived up to its reputation.
In fact, last Wednesday, just purchased a second Lexus; 2008 GX470.

Surprised nobody mentioned about their keys breaking. A constant topic in the threads about replacing keys. Had to have both of mine replaced even though I was very careful about where I hold the key.

forgot to add:
my 07 RX makes this 'rattling' noise for a second after starting the engine. Only happens if I leave the car parked for about 30 minutes when the engine's already hot.
Service Dept says it might have something to do with the VVTi, but 'not to worry' as it is not damaging anything

At 40,000 miles I begin to see moisture in my right headlight assembly. In my job, I travelled a lot and was too busy to take it in right away. I thought it would dry out during the summer. At 55,000 miles the low beam bulb shorted out; probably due to the moisture. Now the Lexus dealer is quoting me $1,000 to replace the entire assembly, because it is now out of warranty. $1,000 for replacing a burned out headlight is just unreasonable. It seems like this is a common problem that Lexus has failed to correct. Not what I expected from them.

Wife has an '05, and we just completed the 60k service @ dealer when the odometer showed 67k or so. Aside from common maintenance items and other smaller misc. things, we had to change out the steering rack @ $1200. Our local dealer never mentioned any internal Lexus "service campaign"? Could this be a Canda thing, or have others had issue with steering rack? It was suppose to be a small leak, but in order to fix it, they had to replace the whole rack. Fortunately, we have the extended warranty where it paid for most of it. Aside from that, it's been a good vehicle.
