Not sure about IS300's rear seal configuration, but typically,...pull the transmission out, clutch & flywheel, change the seal (pull out old one, install new one). Depending on milage, may as well resufrace flywheel, replace clutch, clutch release bearing, install transmission,...you're good to go. You could try a Toyota dealership, they would be less than Lexus.

On my subie, my total was upwards of 800$ for the rear main seal plus an oil change at a Subaru Dealership. But that was after a small engine fire happened while I was watching so they gave me a discount. On a Lexus I have no idea,but I do know that subie dealerships are not as inexpensive like your average chevy,ford,nissan dealers.
