How do I remove the wood trim panel above the glove box in a 2005 Lexus LS 430?

You gotta start at the ac vent. There is a clip in there that you have to get off. Then the rest will snap out

Do I need to take anything off to get to the clip in the vent? is it hard to put the clip back on?

As shown below (Step 22), there are 4 clips that hold the panel in place. Use a screwdriver and pry with slow and steady pressure to release the clips. Be sure to use tape on the screwdriver blade to protect your trims.
