So Im buying a set of aftermarket wheels from another IS250 owner and he thinks that he has the TPMS installed. Is there a way to tell by looking at the valve stem or something?
Also does the year effect it? 09 work with 06?

they go inside the tires and need to be programmed to work with each individual vehicle

Is the seller local? Compare the valve stems to see if they look like the ones on your stock wheels. 09 should work with 06.
