I have been monitoring this car at a local dealer for a long time. He was asking $15,995 fir about a year. It has 72,000 miles and he will sell it to me for $12,000 cash. What should I do?

You're gonna need more details on luxury package contents and mechanical condition. $12k for a 73k '06 IS350 is an amazing price if it's been babied and taken care of well, but if there's a major underlying problem you're going to end up with a massive money drain.
For personal reference, when searching for an IS my last two options were an 09 89k IS250 ($17.5k) and an 06 109k IS350 ($12.8k). I ending up bought the latter (1st owner, babied like crazy) and my friend bought the former. Of course markets vary, so the average prices in your area are going to be different.

Yeah, you'll have to get a list of the options to know. Look for navigation or Mark Levinson etc. You can always check KBB for the market price. If you decide to go ahead from there, ask for a carfax on it. Since this is from a dealer, then he should be able to provide one for you. If that looks good, then take the car for a spin and make sure it runs smoothly up to highway speed and tracks straight. Shouldn't pull left or right when applying the brakes either. You don't have to throw it around corners hard, but do a little "spirited" driving.
If that all works, consider having a mechanic you trust have a look.

Edmunds, which is what I would use over KBB as a buyer, shows a private party value of $11,540, and a dealer retail value of $13,083 for a base IS350 with 72k miles in clean condition.

It's only worth what someone will pay for it and not a dollar more. Make sure everything on it works correctly. Find out the cars history. Has it been taken care of? How many owners? If a car has problems then people usually dump the car (high number of prior owners). Last thing you want is an inexpensive car that turns into a money pit. Sometimes it pays to shop around and pay $1,000-$2,000 more for a car that has an impeccable maintenance history.
It does have low miles but if those are hard miles with a careless owner be prepared to open your wallet. This goes for any make or model of vehicle. What kind of maintenance history does the car have?
Drive this car and other IS350's in your area to familiarize with them. If one car does something out of the ordinary compared to the other cars you know something is up. With all used cars it's BUYERS beware. If you are worried about any issues have the car checked out and get a PPI. If the seller says no to a pre purchase inspection take it as a hint that something is wrong with the car and they are untrustworthy, move on!
We all want to get a great deal on a car. Just make sure that the one you pick out is the right one for you. Take along a friend to view the car. Your friend will not have the emotional attachment and give you a clear unbiased opinion. You should buy a car as a straight up boring business transaction. Look it over and inspect every part of the vehicle. Does it show signs of anything going wrong. Read up on this forum for a couple of days to a weeks before you go looking at the model your interested in. Does the car show any signs of the problems you've researched? You SHOULD know everything about the car before you go out to inspect and test drive. If you don't do your homework and get burned, I'll be the first to say I told you so, then politely tell you it's going to be an expensive life lesson one that may cost you thousands.
