2006 GS300 Owners manual pg 538 states:

"Toyota Genuine Differential Synthetic gear oil
API GL-5 SAE 75W-90 or its equivalent
."

I have the same grade gear oil on my motorcycle but can't get good access to the fill plugs on the two GS differentials so I just had my Lexus dealer do this. They charged $107 for both including the oil and I had a $50 off coupon for service so it ended up costing me only $57.00. (They normally charge $150 but they had a free tech so the total labor for both diffs was less than an hour thus reducing my cost.)

That sounds very reasonable (disregarding your coupon). Your motorcycle takes more oil than just engine oil? Or is it a shaft drive and that's what you're talking about?

Mine's a chain drive and the only oil I need to put in is regular motor oil, which also lubricates the clutch.

My previous Zuki Intruder was a shaft drive as is my dad's Honda Shadow and my brother's Shadow. All of which use the 75w-90. (My new Kawi Vulcan is a belt drive, though.)

Shift drives are awesome... cleaning and adjusting chains is a PITA. I suppose belts are good for the same reason but have their own problems.
