2005 ES330 Passenger Rear Window not going down
Just had the same issue with my car. I could hear a clicking sound when try to lower it from the drivers window window control.
I saw this posted and tries it. I opened the passenger rear door, held the switch down and slammed it shut - IT WORKED!!! Amazing!

I noticed that if you slam the door while holding the switch it would go up, I then got someone to hold the driver switch while I slammed the door - that worked too - most of the time.
I then pulled the panel and narrowed it to the door MPX, I pulled the board out of the casing and narrowed it to the relay, if you tap the relay it goes up every time.
For $30 off eBay I'll just but a used module - but I am sure the relay could just be replaced too.

Exactly, that is how I just diagnosed mine. It is the passenger front, it would sometimes not go up or even go down when you press up.

Sent you a PM about the relay. Please post here to help others if you can, too.

2001 LS430 Driver Window won't go up
My 2001 LS430 driver window started to get stuck in the down position from time to time. I would try it the next day and it would work. I thought it might be a bad switch so I replaced the switch with another used one that I bought but that also did not work. Here is a video on how to remove the switch:
I went to my local dealer and asked about the computer module discussed above. The service adviser reported that he had never seen a bad computer module in 22 years of working there and that the problem was probably a bad window regulator or motor. I replace both the regulator and motor and now the window works just fine. Here is a video on removing the door panel which is the first step:
Finally, here is my video on replacing the regulator and motor:
I hope this helps someone.

GS driver window problems
Thanks for your post.
We just picked up a creampuff 02 GS300 and the
only problem with the entire car was the drivers window acting weird when it was down.
Before I saw your story, I checked the master switch, then the motor, no luck.
Saw your story, bought a used control module from a junk yard for $40.
Hurray!!! Fixed it perfect.
Very easy to work on that door for anybody that would wonder if they should try.
Scottsdale Lexus wanted $300 to look at it.

Sorry for reviving this old thread, but I have this issue on my ‘01 GS300. I read through the thread and bought myself the MPX box off of ebay used. I removed the panel and swapped out the box and tried to use the window on driver’s door which worked, but none of the other windows worked and the cabin light was flashing. Anybody who saw this on their car? If so, what is the fix for this? TIA!
