Yes the 2001 ES300 has a cabin filter, and it is located on the driver's side floor, to the right of the gas pedal. You'll have to pull out some of the carpeting and remove the access door to get to the filter. The instructions are stated in your owner's manual.

The filter location is up under the dash above the gas pedal. you have to pull the carpet back to get at the slots.

Whether or not you have actual filters in place is a different story. Not all owners of pre-2002 ES's have them installed or even know about them. Check your owners manual for instructions (look under air conditioning filter).
