It's possible, how much money you willing to spend? Because it will add up quick, trans, linkage, pedal, brake pedal, slave cylinder, clutch, wiring, etc. My opinion not worth the swap, automatic luxury cars for a reason.

I've look into this casually, one problem for the 99-01 ES300 would be: I am not sure if the VVT-i version is possible to do as a manual for engine control.

When a manual swap is done, whether it is 1MZ-FE into Celica, or 5 spd swap into Camry, the swappers have always used the non-VVT-i version of this motor from the 97-01 Camry. Becuase the Camry had a stock 5-spd available you can get a manual trans ECU to run the motor. Toyota never built a car with the VVT-i version of the 1MZ with a 5 spd, so there is no stock ECU to run it.

Now can you run a 5-spd trans with this motor using the automatic ECU? Maybe, it might work, though I never saw anyone do the VVT-i version swap in reading, and they typically use a 5-spd ECU specifically and you would think there is some reason they don't just keep the automatic ECU. I never looked further than that into it.
