I think the answer here is yes. Even though you don't have the Mark Levinson audio, the "factory" system still has a hidden amp that pushes the sub mounted in your back deck (from what I have learned so far). So you still need this kit, at least that's what I was told!

I'm also thinking about adding navigation to my 2006 ES330, no nav or ML. I don't want to upgrade the sound system or go aftermarket because of the fit.

There are a few OEM units on ebay for a reasonable price. I understand that the ML system is totally incompatible with my Premium/Pioneer setup, but I can't tell if these OEM units are ML or Pioneer. Most are model 86120-33620, Denso.

How can you tell which units are compatible with my premium/Pioneer system?

folks is "Metra TYTO-01 JBL Amplifier Interface Harness" an absolute requirement?

what happens if you put regular wire harness specific for this year toyota/lexus? Will you have problems with an amp?

I am waiting on my last cable and am going to put in a double din GPS Sony unit with rear camera and bluetooth/microphone setup.

Yes, you need that metra kit, otherwise it's not going to work. The ES330 has an amp, on both the factory and mark levinson systems, so your harness must be designed to work with that. A regular harness will not have the smarts to work with an amp on the other end. The TYTO-01 comes with a brain unit that works magic between the head unit and factory amp that power all the factory speakers.

I have 06 ES330 with NAV and Mark Levinson. How much does that change things with what you all have described above? Most people have stated Non-NAV and Non-Mark Levinson.

What I'm looking to do is, at a minimum, change out all speakers and get bluetooth to connect to my Galaxy S4.

Thoughts? Suggestions? Thanks!
